摘 要
植酸酶广泛存在于植物、微生物、动物中,应用前景广阔.本文以黑曲霉400123为原始菌株,采用磷钼蓝比色法测定植酸酶活为指标,以麸皮为基质固体培养基,在单因素试验基础上,应用响应面法优化设计固体培养工艺参数,并对其产植酸酶的酶学性质进行初步研究.结果表明麸皮基质中添加硫酸铵1.24%、料水比1.5、玉米淀粉5.49%、pH 7.26为最优条件,在此优化条件下,28℃恒温培养5d,黑曲霉固体发酵产植酸酶达278.5400U/g,与理论预测值266.9403U/g相符;该酶最适反应温度为40℃,pH为6.5;金属离子Fe2+、Mg2+、Ca2+、Na+一定浓度的添加都表现为对该酶有激活作用,其中最适添加量分别为1.5 mmol/L、5 mmol/L、1.5 mmol/L、0.5,高于9 mmol/L钙离子表现为抑制作用.实验结果为黑曲霉菌产植酸酶的开发及应用提供理论参考.

Abstract
Phytase exists in plants, microorganisms, animals, the application prospect. In this paper, the original strain of Aspergillus niger 400 123, using phosphorus molybdenum blue method for the determination of Phytase Activity Index, wheat bran solid medium as a substrate, in the single-factor test based on the response surface method for optimum design parameters of solid culture and its enzymatic properties of phytase in a preliminary study. The results showed that bran added ammonium sulfate 1.24% matrix material to water ratio of 1.5, corn starch, 5.49%, pH 7.26 as the optimal conditions, the optimized conditions, 28 ℃ incubation 5d, solid fermentation of Aspergillus niger phytase of 278.5400U / g, with the theoretical prediction 266.9403U / g match; enzyme optimum temperature 40 ℃, pH 6.5; metal ions Fe2 +, Mg2 +, Ca2 +, Na + have shown the addition of a certain concentration to activate the enzyme the role of optimal dosage of which were 1.5 mmol / L, 5 mmol / L, 1.5 mmol / L, 0.5, higher than 9 mmol / L calcium ions showed inhibition.
